Dieser Semidünnschnitt zeigt quergestreifte Skelettmuskelzellen, die hellblau angefärbt sind. Das marmorierte Muster im Querschnitt der Skelettmuskulatur ist durch schräg getroffene Zonen der Sarkomere bedingt: Mit 1,5 µm ist ein Sarkomer länger als der Semidünnschnitt mit 1 µm dick ist.
Zwischen den einzelnen Muskelzellen liegt ein zartes, feinfaseriges Bindegewebe, das Endomysium. Eine Anzahl von Muskelzellen wird durch Perimysium internum zusammengefaßt, das ebenfalls aus lockerem Bindegewebe besteht. In das Perimysium internum sind Arteriolen, Venolen und Kapillaren eingelagert. Dicht neben den Arteriolen sind Mastzellen erkennbar, die durch ihr dunkler gefärbtes Zytoplasma auffallen.

Richardson stain

Semi-thin sections are often stained by Richardson technique with a blue dye, Methylene blue Azure II.

Basophilic and osmiophilic structures stain blue, and metachromatic structures turn red-violet.

Semi-thin sections of tissue specimens are generally prepared for the light microscopic examination before ultra-thin sections are prepared for examination under the transmission electron microscope (TEM). Semi-thin sections are thinner than other sections commonly used for light microscopy.

The embedding agent for TEM examination is a plastic resin, which is harder than paraffin. Since plastic resin cannot be detached easily and without creating artifacts, in contrast to paraffin, there exist only few suitable staining techniques for tissue embedded in plastic resin. Richardson stain is one of them.

Mastzellen kommen, einzeln oder in Grüppchen, am häufigsten in der Adventitia kleinerer Blutgefäße vor, sind aber auch im lockeren Bindegewebe und Stroma verschiedener Organe weit verbreitet. Bevorzugte Organe sind: Lunge, Haut, Magen, Darm, Leber- und Milzkapsel. Mastzellen sind langlebig und können sich im Gewebe vermehren.

You should then decide what structure this could be. To check your result, simply click the appropriate square, and the correct label will appear. The option “marked” allows you to see all labels for all structures simultaneously. These can be removed by clicking “unmarked”. This reactivates the dynamic labels.
