Die quer getroffenen Skelettmuskelfasern liegen dicht aneinander und erscheinen meist hexagonal. Direkt unter dem Sarkolemm liegen die Zellkerne. Nicht immer sind diese leicht von den extrazellulär, in feinem umgebenden Bindegewebe liegenden Fibrozytenkernen zu unterscheiden. In der unteren Hälfte ist der Anschnitt einer Bindegewebssepte zu sehen, die mehrere Muskelfasern zu einem Bündel zusammenfaßt. In diesen Bindegewebsscheiden (Perimysium) laufen die Nerven und Gefäße. Die weißen Risse innerhalb der Muskelzellen sind Schneideartefakte. Die feine Punktierung des Querschnittbildes entspricht den quergetroffenen Myofibrillen.

Hematoxylin and Eosin (H&E)

H&E is the most common survey stain, which shows cell nuclei and cytoplasmatic components.

Dyes:

Hematoxylin (e.g. hemalaun, Weigerts or Heidenhains iron-hematoxylin)
Hematoxylin is a positively charged dye (at low pH values). It therefore colors negatively charged (basophilic) structures blue, like the DNA of nuclear chromatin, the RNA of rough endoplasmatic reticulum, and the acidic glycosaminoglycans of hyaline cartilage.
Hematoxylin used in the preparation of iron varnish is Weigerts or Heidenhains iron-hematoxylin and in the preparation of aluminum varnish is hemalaun.
Eosin Eosin is a negatively charged dye used for counterstaining. It stains all remaining structures different shades of red (acidophilic).

Thiazinrot

Thiazinrot behaves similar as eosin.

Jede einzelne Muskelfaser wird von einer zarten Bindegewebsschicht, dem Endomysium, umhüllt. Das Perimysium internum umscheidet mehrere Muskelfasern und faßt sie zu einem Primärbündel zusammen. Mehrere Primärbündel werden vom Perimysium externum umgeben und bilden ein Sekundärbündel. Das Epimysium umschließt den ganzen Muskel, der durch das Perimysium in Septen geteilt ist. In den aus überwiegend kollagenen Fasern bestehenden Septen verlaufen die Gefäße und Nerven.
Die Innervation der Skelettmuskulatur erfolgt willkürlich über myoneuronale Verbindungen (motorische Endplatten). Jede Muskelfaser steht über mindestens eine motorische Endplatte mit dem peripheren Nervensystem in Verbindung.
