Die stärkere Vergrößerung der Milz zeigt, daß die dunkler gefärbten Areale durch knötchenartige Anhäufungen von Lymphozyten zustande kommen. Sie werden Milzknötchen oder Malpighi-Körperchen genannt und gehören zur weißen Pulpa. Zum Teil zeigen sie einen helleren Innenbereich, das Keimzentrum. Die heller angefärbten und locker stukturierten Areale gehören der roten Milzpulpa an, die von Sinusoiden durchsetzt ist. Die nicht angefärbten Bezirke stellen die Lumina der Sinusoide dar. Die Makrophagen sind als dunkle Granulahäufchen erkennbar. Die schwarzen Einlagerungen sind die in vivo eingespritzten Tuschepartikel, die von den Makrophagen phagozytiert und gespeichert wurden. Makrophagen finden sich im Grenzbereich zwischen weißer und roter Milzpulpa, der sog. Marginalzone und nahe den Sinusoidwänden der roten Pulpa.

Monocytes

Monocytes are part of the immune system – the mononuclear phagocyte system. Chemotactic stimuli cause them to cross the capillary walls and enter the connective tissues, where they differentiate into macrophages. Here they can be found in both mobile and sessile (histiocytes) forms.

Hematoxylin and Eosin (H&E)

H&E is the most common survey stain, which shows cell nuclei and cytoplasmatic components.

Dyes:

Hematoxylin (e.g. hemalaun, Weigerts or Heidenhains iron-hematoxylin)
Hematoxylin is a positively charged dye (at low pH values). It therefore colors negatively charged (basophilic) structures blue, like the DNA of nuclear chromatin, the RNA of rough endoplasmatic reticulum, and the acidic glycosaminoglycans of hyaline cartilage.
Hematoxylin used in the preparation of iron varnish is Weigerts or Heidenhains iron-hematoxylin and in the preparation of aluminum varnish is hemalaun.
Eosin Eosin is a negatively charged dye used for counterstaining. It stains all remaining structures different shades of red (acidophilic).

Thiazinrot

Thiazinrot behaves similar as eosin.
Makrophagen kommen in der Milz vor allem an zwei Stellen vor, Die Makrophagen der Marginalzone stehen im Dienst der Infektionsabwehr. Hier findet die erste Auseinandersetzung mit dem Antigen statt.
Die Makrophagen im Maschenwerk der roten Pulpa beseitigen überalterte rote Blutkörperchen.
